Avoiding barriers to PET radioligand development: cellular assays of brain efflux transporters.

چکیده
PET provides a uniquely sensitive and powerful means for studying a clinically relevant protein target within the brain, provided that an adequately specific and sensitive radiotracer is available. Interesting targets, which include neurotransmitter receptors, neurotransmitter transporters, enzymes, and plaques, far exceed the availability of effective radiotracers. Most PET radiotracers are designed to bind avidly and reversibly with their target. This type of radiotracer is usually termed a radioligand. Candidate radioligands for imaging must ideally satisfy an array of demanding criteria. These include ability to penetrate the blood–

Benzylamine (4-18F-ADAM): An Improved PET Radioligand for Serotonin Transporters
There has been considerable interest in the development of PET radioligands that are useful for imaging serotonin transporter (SERT) in the living human brain. For the last decade, 11C( )McN5652 has been the most promising PET agent for studying SERT in humans. However, this agent has some limitations. Recently, a new promising SERT PET radioligand, 3-11C-amino4-(2-dimethylaminomethylphenylsulf...
متن کاملMultidrug resistance-associated proteins: expression and function in the central nervous system.
Drug delivery to the brain is highly restricted, since compounds must cross a series of structural and metabolic barriers to reach their final destination, often a cellular compartment such as neurons, microglia, or astrocytes. The primary barriers to the central nervous system are the blood-brain and blood-cerebrospinal fluid barriers. Through structural modifications, including the presence o...
متن کاملIn vivo imaging of vesicular monoamine transporters in human brain using [11C]tetrabenazine and positron emission tomography.
The pharmacokinetics of [11C]tetrabenazine, a high-affinity radioligand for the monoamine vesicular transporter, were determined in living human brain using in vivo imaging by positron emission tomography (PET). The radiotracer showed high brain uptake and rapid washout from all brain regions with relatively slower clearance from regions of highest concentrations of monoamine vesicular transpor...
متن کاملFactors That Limit Positron Emission Tomography Imaging of P-Glycoprotein Density at the Blood–Brain Barrier
Efflux transporters located at the blood-brain barrier, such as P-glycoprotein (P-gp) and breast cancer resistance protein (BCRP), regulate the passage of many drugs in and out of the brain. Changes in the function and density of these proteins, in particular P-gp, may play a role in several neurological disorders. Several radioligands have been developed for measuring P-gp function at the bloo...
متن کاملAdvances in the Development of PET Ligands Targeting Histone Deacetylases for the Assessment of Neurodegenerative Diseases.
Epigenetic alterations of gene expression have emerged as a key factor in several neurodegenerative diseases. In particular, inhibitors targeting histone deacetylases (HDACs), which are enzymes responsible for deacetylation of histones and other proteins, show therapeutic effects in animal neurodegenerative disease models. However, the details of the interaction between changes in HDAC levels i...
